To determine if Bs20x22 maintained similar binding characteristics as the parent mAbs, Ramos cells (CD20/CD22 positive) and 293T cells transfected with either CD20 or CD22 were stained with Bs20x22, rituximab, or HB22.7.
Animals received one of the following antibodies by intraperitoneal injection: 6G1, which is pan-amyloid-β specific and has similar binding characteristics as the diagnostic 6E10 antibody (Barghorn et al., 2005); A-887755, a globulomer-derived antibody which specifically detects oligomeric, but not monomeric or fibrillar amyloid-β (Hillen et al., 2010); IgG2a as a non-amyloid specific control.
Notably, even such fundamental binding characteristics as the equilibrium dissociation constant (Kd) between Nups and NTRs are still matter of discussion - estimates range from a few nM to several mM (Bednenko et al., 2003; Ben-Efraim and Gerace, 2001; Tetenbaum-Novatt et al., 2012; Tu et al., 2013).
