It is likely that this 7 kb sequence at the right end represents the start of the right TIR, and was acquired by some of the long PacBio reads that extended from the region next to the TIR, while the rest of the reads from the right TIR have probably been assembled with the left TIR.
Moreover, we found that Crk SH2 domain binds Tir through Y474 and competes with the binding of the Nck SH2 domain to Tir, thus inhibiting its recruitment and subsequently, N-WASP activation.
The successful synthesis of TiR and FTiR on a FTO glass substrate can be seen in the SEM and TEM micrograms shown in Figure 2. Figure 2a shows the SEM image of the top of the TiR film, while the insert shows the cross-sectional view.
We wondered whether the binding of Tir to cortactin would activate the latter and promote Arp2/3 complex-dependent actin polymerization.
We propose that cortactin binds Tir through its N-terminal part in a tyrosine and serine phosphorylation independent manner while SH3 domain binding and activation of N-WASP is regulated by tyrosine and serine mediated phosphorylation of cortactin.
The binding of intimin to Tir mediates the adhesion between the pathogen and its host cell[19].
Shortly after the successful binding, the translocated Tir protein triggers additional signal transduction and actin polarization in host cells, which are essential for lesion formation[20], [21], [22].
Tc1/mariner family transposon termini comprise at least three types of functional sequences involved in transposition: the 4 7 nucleotide TE cleavage sites at the outer extremities of the TIRs, the DRs within the TIRs, which are the transposase binding sites, and the UTRs, between the TIRs and ORFs, which act as enhancers of transposition efficiency [ 49, 57- 59].
In the six examples shown here, the extreme outer region of the TIRs is maintained while the inner region of the TIRs is not present.
These findings provide structural insight into the binding pattern of intimin to Tir and the molecular mechanism of EHEC O157: H7.
