We considered these 24 known targets as "true positives" for the phenotypes of interest in one of the benchmarks described below (see section Validation of predicted therapeutic targets in Results and discussion).
Lastly, a spontaneous tandem duplication within Fbn1 is responsible for the phenotype of tight skin (Tsk/+) mice, which includes myocardial, skeletal and pulmonary abnormalities of MFS and fibrotic features of SSS (see below) [ 43].
The first phase of phenotyping of the animal model should be complemented with systematic observations (and eventually, specific tests for detecting sensorimotor dysfunctions) that allow monitoring and assessing the welfare of the model animal [ 23, 121]; see below).
Western blot analysis confirmed that embryos produced by either method lacked detectable PLP (Fig. 3A; data not shown), and both sets of embryos exhibited the same phenotypes that we describe below; for ease of presentation we simply refer to these embryos collectively as plp mutants (plp mut).
