A fragment containing the rpoB-promoter region, the rpoB-leader, and the Shine-Dalgarno and startcodon was PCR amplified using the primers Prpofwd and PrpoRev2 (5'-GCGGATCCTGCCAAACAGATTCAC-3'), digested with EcoRI and BamHI and ligated into pLUG520 where the PrpoB-spa-fragment had been removed using the same enzymes.
