The regions of interest (beating area, immunostaining area) were defined in Photoshop (Adobe systems) using the 'magic wand' tool.

We noted that while all three CMs increased the percentage of EBs that formed beating areas, beating areas remained relatively small in response to END2-CM.
While embryoid bodies transfected with no siRNA or a scrambled sequence had normal beating areas with high electrical activity (red) (Fig. 6C), embryoid bodies transfected with triple siRNA developed sparse and poorly organized beating areas if at all (Fig. 6D).
