Residual competition from partially C- or N-truncated peptides was observed, implying that some of the aa positions flanking the 3 10 helix may be dispensable for binding.
The IG1 and IG2 of ROBO is essential for the Slit Robo interaction [ 24]. Biochemical studies show that the interaction between SLIT and ROBO involves the highly conserved second LRR domain (D2) of SLIT and the IG1 domain of ROBO, while IG2 IG5 and all FN3 domains of ROBO1 appear to be dispensable for binding [ 16, 23, 25, 26].
As we had already established that the FKH domain was dispensable for binding to c-Rel, we based the various deletion and point mutants on Foxp3ΔFKH (Fig. 2).
