High glucose significantly increased basal l-arginine transport 2.1 ± 0.4-fold (Fig. 2A).
Basal l-arginine transport was significantly increased under high glucose culture conditions, yet insulin-stimulated l-arginine transport remained unaltered.
Examination of an association between basal L-selectin expression and ESR in patients with RA revealed a negative correlation between these variables (R2 = 0.29, p < 0.05; Figure 2b).
There was roughly 5-fold lower basal L-βHB dehydrogenase activity than D-βHB dehydrogenase activity in the worm extracts (data not shown).
However, correlative analysis of ESR and basal L-selectin on monocytes revealed a similar trend of negative correlation to that for neutrophils (R2 = 0.29, p = 0.06; Figure 2c).
Interestingly, knockdown of dhs-2, dhs-16, or dhs-20 decreased basal L-βHB dehydrogenase activity in the extracts (Supplementary Figure 2).
In HAECs infected with Ad.Akt-CA, basal l-arginine transport was significantly increased by 44% compared to control adenovirus infected HAECs, yet insulin stimulation had no further significant effect on l-arginine transport (Fig. 1C).
The glucose-stimulated increase in basal l-arginine transport was unaffected by preincubation with wortmannin, but was abrogated by incubation with the MEK inhibitor, PD98059, or the PKC inhibitor, bisindolylmaleimide I (Fig. 2B).
Relative to CD44v6 expression, the probability of recurrence was 94 and 75% for cases that showed basal (L), or basal and middle (M) staining, respectively, and 38% for patient that had full-thickness (F) positivity (two-sided log-rank test, P=0.007; Figure 4B).
The strength of differential expression between any pair of experiments was estimated by M i = log2 ratio) = X i- X a, where a represented one particular cell type and i represented each given cell type in the set: B (basal), L (luminal), S (stromal), E (endothelial), G3 (Gleason 3) or G4 (Gleason 4).
