Since Cortactin is relatively more abundant than c-Src (20 times more whole cell lysate is required to detect c-Src compared to Cortactin), we hypothesized that the increase in the levels of phospho-Cortactin (Tyr421) we observed in Tbdn knockdown cells may be sufficient to stabilize activated c-Src and increase the basal levels of activity of c-Src.
