Products of ∼200 ± 25 bp (insert plus adaptor and PCR primer sequences) were band isolated from an agarose gel, purified using Gel Extraction kit (Qiagen, Hercules, CA), and PCR amplified using Phusion Polymerase with Illumina primers for 18 cycles.
DNA was separated by agarose gel electrophoresis, one band isolated and the agarose fragments containing the DNA divided in half.
