Xenogenic DNA vaccination against the melanoma differentiation antigens glycoprotein (gp) 100 [ 6- 9] and tyrosinase (tyr) [ 10- 13] have been shown to overcome auto-tolerance and to elicit an immune response in mice, dogs, and humans [ 10- 16] and a clinical antitumoral effect in mice and dogs [ 10- 14, 16, 16].
To address the relationship between inflammation and neurodegeneration we used an auto-immune tolerance strategy to eliminate clinical relapses in EAE in a manner analogous to the clinical effect of disease modifying treatments.
IgG immuno-intensity, used as a surrogate of general immune-reactivity noting its association with microglia [ 26, 27] and blood brain permeability [ 21, 28, 29], was examined to further confirm the effectiveness of auto-immune tolerance to halt immune reactivity.

Uniquely, pre-naïve B cells expressed CD5, a marker of the B-1 subset of murine B cells, which can function as a negative regulator of BCR signaling and promote maintenance of tolerance to auto-Ag [ 15– 17].
High doses of vitamin D3 leading to supraphysiologic range of circulating 25(OH)D3 may compensate for this genetic-related status of relative vitamin D deficiency, thereby establishing tolerance to auto-antigens.
