We initiated a culture by growing a single colony overnight in 10 ml LC medium with 120 rpm shaking, to attain stationary-phase density (PP: ~4 × 10 cells ml−1; ERA: ~5 × 10 cells ml−1).
This is in accordance with the force attained in phase 2 (T2) following step length perturbations in fibre experiments28,29 (Supplementary Fig. 6b).
