As the ameloblasts progress to maturation, the migrating SI cells increase in size and adopt a stellate appearance.
During the pre-secretory stage, ameloblasts are separated from the adjacent neural crest cell-derived odontoblasts by a basement membrane which is subsequently removed as the ameloblasts enter their secretory phase.
As the ameloblasts retreat away from the dentine, they lay down an extracellular matrix within which the hydroxyapatite crystals begin to form.
As the ameloblasts entered the maturation stage, during which rapid crystal growth appears and mineral content dramatically increases, they shortened and the proteins were removed from the enamel ECM as evidenced by the loss of staining (Fig. 4C).
As the ameloblasts enter their maturation phase, the organic matrix is degraded completely to allow secondary growth of the hydroxyapatite crystals which ultimately occlude the spaces previously occupied by the enamel extracellular matrix proteins.
In later stages of teeth development, this area extended as the ameloblast differentiation progressed and proliferation decreased in teeth mesenchyme (Fig. 6d).

(4) During the secretory stage of amelogenesis, long thin ribbons of enamel mineral crystals are formed almost immediately as the ameloblast lays down enamel matrix proteins, suggesting that enamel mineralization does not take place within a preformed matrix.
Most significantly, human keratinocytes, recombined with mouse embryonic dental mesenchyme in the presence of exogenous FGF8, are induced to express the dental epithelial marker PITX2 and differentiate into enamel-secreting ameloblasts that develop a human mouse chimeric whole tooth crown.
Ameloblastin continued to be detected within the ameloblasts as maturation proceeded, with relatively little staining present in the underlying enamel matrix (Fig. 5J L).
