B31 and its related species, B. afzelii PKo and Borrelia garinii PB, have been sequenced, as well as many of the associated plasmids [ 20- 23].
High quality sequence data has opened the possibility of systematic dissection of how genes distributed on both the bacterial chromosome and associated plasmids have made it such a successful pathogen.
This flagellin gene has been mentioned in previous CNT studies due to its close proximity to the neurotoxin gene cluster [ 13] and its existence in numerous C. botulinum type A strains and associated plasmids [ 25].
We used three activity assays including HeLa cell adhesion, human blood hemagglutination and bacterial clumping tests and polymerase chain reaction (PCR) primers designed from an aggregative adherence pattern associated plasmid (pCVD432) to screen the EAggEC strains in 403 E. coli isolates including 63 laboratory isolates and 340 clinical isolates obtained from diarrheal disease cases.
EvoPrinter analysis of the MRSA252 chromosome also uncovered virulence-factor encoding HGT events with the genome of Listeria monocytogenes and a Staphylococcus saprophyticus associated plasmid.
Thus, these results confirm the presence of at least five phylogenetically distinct subgroups of IncP-1 plasmids and represent the first time that sequences associated with plasmids of all of these five subgroups have been detected in a single setting.
Some of the larger-scale genomic differences among the four Pae strains appeared to be associated with plasmids and all four strains varied in their plasmid complement.
In addition, the fact that all but the island containing the avrGf1 gene contain genes that are normally associated with plasmids (ccdB, mobL, cvaB) indicates a likely xenologous plasmid origin.
Under this evolutionary model the plasticity of plasmids can be explained in relation to life history and overall fitness costs associated with plasmids: those lineages with strict host specialization, particularly lineages undergoing extreme gene loss and accelerated rates of nucleotide evolution (i.e., TG rickettsiae), would no longer need to harbor plasmids.
