On the contrary, but consistent with the lack of WAT browning in VEGFAdKO-HFD-IF mice, loss of adipose-VEGF completely eliminated the IF-mediated increase of alternatively activated adipose macrophage (Figure 6B; Supplementary information, Figure S10B and S10C).
Immunohistochemical analysis of SAA3 protein revealed intense staining of epididymal adipose tissue (Figure 1C) with minimal background in treatments omitting the primary SAA3 antibody (Figure 1D).
The obese and lean TRAP over-expressing lines both expressed elevated levels of TRAP mRNA (WT vs TRAP+ Z = 2.94 p = 0.003) in adipose tissue (Figure 2 E), so the basis of differential adiposity is not due to differential expression of the transgene.
In contrast, BMP-6 was only expressed after one week of implantation in the inflammatory and adipose tissue (Figure 1).
Histological examination showed that the tumor consisted of many epithelioid myoid cells, epithelioid clear cells with sinusoidal vessels, and mature adipose tissues (Figure 4B).
As expected, high uptake of [18F]FDG was also observed in the heart, bladder, brain, kidneys and brown adipose tissue (Figure 3a).
This is in agreement with our observations in mice where ZEB1 increases in response to increased adipose accumulation (Figure 1).
Spred1 is also more highly expressed in both adipose tissue (Figure 10E) and liver of WKY animals.
Data for the cell perimeter of white adipose tissue (Figure S2) was analyzed with Mann-Whitney U test.
