Nonetheless, in most reports the additional lifespan of the post-senescent cell lines expressing SV40LT is still restricted by another barrier called crisis.
One clue comes from the observation that calorie-restricted long-lived flies do not show additional lifespan extension when DN-Dmp53 is also expressed, indicating that Dmp53 and calorie restriction are related (Bauer et al., 2005).
MPK-1/ERK phosphorylates SKN-1, promotits its activation and nuclear accumulation, and knockdown of MPK-1/ERK by RNAi does not result in additional lifespan reduction in the absence of functional SKN-1, suggesting that Ras/ERK signaling can promote lifespan extension, at least in part, through activation of SKN-1 and the antioxidant response [ 50].
We observed that oga-1 mutant lifespan was significantly longer than that of wild type (three independent experiments showed significant differences; p<0.0001, n=66-89 for each experiment); and double mutant analysis of oga-1 with each of three separate components of the insulin signal transduction pathway, daf-2, age-1, and sgk-1, did not show additional lifespan increases in our experiments.
Insights into the mechanisms of this neuronal Dmp53 effect were provided by the finding that calorie restriction (CR) of the neuronal DN Dmp53 flies does not provide any additional lifespan extension beyond that observed in non-restricted DN Dmp53 flies [ 17].

Lifespan as a measure of fitness is OK, but the effect size shown here (i.e., 4 8 days of additional life) seems quite modest.
For a total of 560 exposed individuals, we report measurements of four life history traits: age at first reproduction, size (body length measured for each individual on day 12, see Additional file 1), lifespan and lifetime number of offspring.
Although worms cultured at 25°C had reduced total reproduction and lifespan (Additional file 1: Table S2) compared to those cultured at 20°C (Additional file 1: Table S1), their strong early-life reproduction allowed them to achieve similar levels of relative fitness (compare Figure 2H and G).
Maximal lifespan (Additional file 1: Tables S1 and S2) showed the opposite trend for both assays.
As predicted, they do not experience additional increase in lifespan compared to esgGal UAS-dPGC-1 (Fig. 5A,B, S5A, TablesgGal UAS-dPGC-1
