The global activator GacA up–regulates the receptors LasR and RhlR of the first and second QS system, whereas the activator Vfr only up–regulates LasR [ 12, 13].
To conclude our analysis, we observe rpmC as down-regulated, however, its inhibitor is down-regulated and its activator is up-regulated; we should conclude that rpmC is up-regulated, yet its observed change tells us the opposite, therefore, we find an inconsistency between model and data (Fig. 2B).
xlnR (AN7610), which encodes the xylanolytic/cellulolytic transcriptional activator, was up-regulated at the last time point (consistent with creA decrease [ 98]), notwithstanding the up-regulation of the xlnA and xlnB (AN9365) at the earlier time points.
This suggests that a critical role of Mediator in vivo is to help activators open up the chromatin structure in order to enable the general transcription machinery to gain access to the core promoter.
In line with its signal transduction mechanism as a transcriptional activator, activated Notch1 mostly up-regulated, rather than down-regulated, the expression of target genes (Fig. 2C).
This result can be explained if we assume that STATc acts together with a transcriptional activator for the up-regulation and/or a transcriptional repressor for the down-regulation of target genes (see Fig. 6).
It is important to note that a possible role of PPARα cannot be excluded because specific PPARα co-activators or up-stream factors were down-regulated.
These results may not only reveal equally dynamic changes in abundance for both the increases and decreases in the total gene expression for different categories in resistant house flies, but also indicate an important feature of resistance gene regulation by both activators (the up-regulated genes) and perhaps, repressors (those down regulated genes).
Instead, our results demonstrate that the apparent SRS function of Tat is directly related to its activity as a transcriptional activator, and the up-regulation of silencing reporters can be accounted for by their increased transcription, as argued previously [14].
