The finding of these two activating modifications to be the strongest in telomeres is interesting and is likely to be related to the recent finding of RNA polymerase II transcription at mouse[ 34] and human[ 35] telomeres.
Recent investigations have also suggested that 5-aza has broader effects than purely as a demethylating agent, including altering activating histone modifications to repressive marks, and expression changes independent of promoter methylation (Komashko et al, 2008).
This leads to chromatin remodeling and changes in histone modifications to activate and repress target gene transcription [ 1– 3].
Taken together, we illustrate approaches that can be taken to analyze large genomic data, and provide evidence that NFI family members may act in conjunction with specific chromatin modifications to activate gene expression.
