The majority of these drugs are currently produced in altered Chinese hamster ovary (CHO) cells due to the high viability and the high densities achieved by these cells in suspension cultures.
With the small molecule-aided episomal reprogramming method, high reprogramming efficiency was also achieved with these cells (∼800 iPSC colonies from 1×106 input post-expansion cord blood cells, i.e. ∼600 iPSC colonies per 1×104 input freshly isolated cord blood CD34+ cells with 75-fold expansion during 7-day culture) (Fig. 4C).
Of interest, lower induction of IFN-induced genes in response to dsRNA was observed in two of the most permissive cell lines, Huh7-2 and Huh7-9 suggesthat that defects in dsRNA signaling may be contributing to the high HCV levels achieved in these cells.
