In order to confirm the involvement of TLR2, we transfected NCI-H292 cells with siRNA against TLR2 and found that such inhibition resulted in abrogation of effect of PGN on epithelial wound healing (Fig. 4A).
If the regulatory mechanisms present in yeast and mammals also are used by T. gondii, disruption of critical RPs would lead to abrogation of effect of an MDM2-like molecule, arrest in the cell cycle in G1, and hence contribute to diminution of proliferation and persistence.
Although our data are of a preliminary nature, we note that in MCF7 cells with ZNF703 overexpression treated with TGFβ, there was less activation of the pathway as measured by RT-qPCR of target genes (TGFBI and PAI1), in association with abrogation of effect on proliferation (Fig 5B).
This novel finding on the abrogation of effects of disease activity on JSN by combination therapy with ADA+MTX appears to be of particular importance in light of recent observations that JSN may affect physical function more profoundly than bony joint damage.
It is possible that either through abrogation of the effect of E7 or as a result of epigenetic or mutational silencing of host p16 (O'Regan et al, 2008), the HPV+ve/p16−ve tumours lack the molecular consequences of E7 expression and that one of the consequences of this is lower radiosensitivity compared with HPV+ve/p16+ve tumours.
