The phrase "a supposition that" is correct and usable in written English.
It can be used when discussing an assumption or hypothesis that is being considered or proposed.
Example: "The study is based on a supposition that increased exercise leads to improved mental health."
Alternatives: "an assumption that" or "a hypothesis that".
