The phrase "a significant defect" is correct and usable in written English.
It can be used when discussing a notable flaw or issue in a product, process, or system that has considerable implications.
Example: "The quality control report highlighted a significant defect in the manufacturing process that could lead to product failures."
Alternatives: "a major flaw" or "a serious issue."
