Virus production in culture supernatants showed an increase generally at 7 days PI (Figure 2), although some cultures showed the opposite trend with a replication peak at 72 h PI in culture supernatants.

In order to estimate replication peak resolution, we amplified, cloned in a URA3-plasmid, and transformed in C. glabrata, eight regions of 4 kb surrounding origins in which no ARS was detected.
With extensive smoothing (0.075 span value), close replication peaks tended to merge into one single peak, like, for example, the large left-most double (or triple) replication peak.
However, using T6 and T0 coverage, which correspond respectively to S and G1 phases of the cell cycle, we were able to determine replication peak positions.
Although the position of NUMTs at previously identified replication peaks is intriguing, it may not be a direct indication of the firing activity, since NUMT14 that also displays late-firing activity, is located at a valley.
According to this data, most but not all ARS are located at replication peaks, while some ARS are located at valleys and others at intermediary positions.
The average distance between bona fide replication peaks found in both experiments was 6.9 ± 2.4 kb (95%% confidence interval), consistent with what was deduced from comparisons between ARS positions and replication peaks (Fig. 5a ).
