We used mRNA containing the AUG initiation codon as a positive control and compared it with mRNAs containing a CUG codon in an "Excellent" (UCGACCCUGA) versus a "Poor" context (GCGUCCCUGA).
The first AUG of mammalian RNase ZL is in a poor context for initiation and thus apparently passed by most scanning ribosomes, while the second AUG more closely conforms to a consensus initiation site and appears to be used preferentially.
The first AUG of mammalian RNases ZL is consistently found in a poor context for initiation and is apparently frequently passed by scanning ribosomes.
The PIC can adopt an 'open' state, which makes it easier to scan the mRNA for the correct start codon and ignore triplets similar in sequence to AUG (like UUG) or AUG triplets in a poor context.

Because developing countries generally have a poorer context than developed countries, they confront many challenges in e-government implementation.
Changing the CUG context to a "Poor Kozak" context diminished the level of antigen presentation to almost that of the CCC[YL8] background.
However, there are exceptions: as a result of poor context (for example, noise), the first ATG could be ignored.
Compared to the strong toeprint band at +15 17 nucleotides with the AUG codon and a weaker band with the CUG codon in an "Excellent Kozak" context, a toeprint band was not detected when the CUG was in a "Poor Kozak" context (Fig. 2A, CUG Poor).
Furthermore, given the resource poor context, taking on a stewardship role might prove not to be beneficial as this could have negative financial implications.
Could it be possible that a general decrease in frequency of start codon recognition could just have a much greater effect on the weaker or poor context AUGs and relatively less on the AUG in a good sequence context?
However, even in the case of a gene with strong selection pressure (see Sharp et al. 45 for a discussion of selection pressure on codon usage), the first anticodon position will be a C in a GC-poor context.

Since I tried Ludwig back in 2017, I have been constantly using it in both editing and translation. Ever since, I suggest it to my translators at ProSciEditing.
