Oligonucleotide parameters for Primer3 were set to a length of 18 – 27 bp with an optimum of 20 bp, a GC content of 20 – 80 % with an optimum of 50%%, a melting temperature (Tm) of 57 63°C°C with an optimum of 60°C, and a primer Tm maximum difference of 1°C.
The main primer design parameters were set as follows: PCR products ranging from 100 to 300 nt, primer lengths ranging from 18 to 24 nt (with an optimum of 20 nt), a 60°C optimal annealing temperature, and a GC content from 40%to65%5% (with an optimum of 50%) [ 48].
Group I (proteolytic) C. botulinum is a mesophilic bacterium with a minimum growth temperature of 10-12°C 10-12°Cimum of 37°C and maximum groptimummperature of around 48°C.
Figure 2b shows that A. resinae grew well in a wide pH range with an optimum of pH 4-6.
A subsequent screen of pH values gave an optimum of 7.5.
