The mycobacterial cell wall is composed of a mycolyl arabinogalactan peptidoglycan (mAGP) complex (Daffé et al., 1990; McNeil et al., 1990 , 1991 Besra et al., 1995; Brennan, 2003; Dover et al., 2004), of which the mycolic acids and extractable lipids form the mycobacterial outer membrane (Hoffmann et al., 2008).
A Mutually Attracted Gene Pair (MAGP, see Results) could be composed of two genes in some highly conserved operon, or be a pair of genes conservatively close together irrespective of operon structures.
Furthermore, a decline in MAGP-1 expression with age was observed both at the gene level and at the protein level.
A mature mycolic acid is eventually transferred either to arabinogalactan (to form mAGP) in part by the Ag85 proteins, or to an α,α-trehalose-6-phosphate (T6P, leading to TMM) by an unknown mycolyltransferase [9].
This showed that 618 (58%) MAGP were composed of genes belonging to the same functional category, among which 391 were MAGP maintained by operons.
Later, one of the polysaccharides was identified as arabinogalactan (AG), the basic constituent of the mAGP complex (Misaki & Yukawa, 1966), and the other as a pool of immunologically active lipoarabinomannan and inactive lipomannan (Misaki et al., 1977), both sharing a similar mannan core (Hunter et al., 1986; Hunter & Brennan, 1990; Chatterjee et al., 1992a;) (Fig. 2).
When putting aside the operon effect, if this new MA showed that the two genes still had a very strong attraction (3 sigma larger than the mean of MA drawn from pairs of genes randomly distributed; the mean and standard deviation were from the large class following a normal distribution, retrieved from Figure 7, see Results), we then concluded that this MAGP was not due to operons.
