We first checked by RT-PCR for ovarian expression of Yp genes; nine Yp genes were expressed in XY Sry- ovaries and, among these, Rbmy was clearly reduced in XYd-1 ovaries (Fig. 4A).
The cell samples stored at −80°C were retrieved and diluted with sterile milliQ water to read an OD650 of 0.1 and a volume of 100 μl was spread on three YP-hydrolysate plates.
