Another two proteins, SCAP and INSIGs, including INSIG1 and INSIG2, are critical for the transportation of SREBPs from ER to Golgi apparatus [ 5].
Two Tpi-sCAP transcripts were identified that both encode precursor proteins of 136 amino acids, including spatial conservation of putative cleavage sites and cysteine residues.
However, when we conducted a GMDR analysis, we found the significant interaction of the three genes (INSIG2 rSCAP8762, SCAP rs12487736, and SREBP2 rs1883205) on obesity.
We then performed TR4 ChIP experiments for the other cell types and used standard PCR to confirm enrichment at three sites (TNFIAP1, SCAP, ECSIT) previously identified in HepG2 cells (see Additional file 2 for primer information; see Additional file 3 for representative PCR validation).
