Finally, the AGL080C disruption module was obtained by EcoRI digestion and was used to transform either spores of the A. gossypii ATCC 10895 strain, for the single disruption, or spores of the A. gossypii Δ agr371c strain, for the double disruption.
Because analysis of the isolated N-terminal module indicated possible disruption of its native structure, we also generated and purified the CBS-1 mutants K421A, which abolishes a canonical PLP-binding site in the C-terminal module, and E62K, which creates a putative PLP-binding site in the N-terminal module.
In other words, molecular networks are networks of networks that can tolerate disruptions to individual modules but whose functions are sensitive to disruptions module of connectors.
We have designed a food-safe-grade module for gene disruptions in commercial baker's yeast strains, which contains the G418 resistance cassette, KanMX4, flanked by direct repeats from the MEL1 gene of Saccharomyces cerevisiae.
With the disruption modules A. gossypii ATCC 10895 spores were transformed by electroporation as previously described [ 25].
The final disruption modules comprised the G418 r or Hyg r selection cassettes flanked by the AgPRS2,4 or AgPRS3 sequences, respectively, to enable homologous recombination and genomic insertion [ 30] (Fig. 3).
As disruption of TA modules is a potential drug target, a search for homologous two-member TA modules was made in all the strains of the pathogenic organisms.
Rather, changes occur in a hierarchy of gene modules which collectively contribute to disruption of essential cellular functions [ 15- 17].
