The cell was bulk loaded with Fluo-4 and electrophysiological recordings were performed in the cell-attached recording mode from visually targeted neurons.
To ensure that whole-cell recorded firing patterns were also quantitatively similar to cell-attached recorded firing patterns, we compared the frequency of regular tonic firing in a subset of 33 neurons (P16 P23) that were recorded both in cell-attached (before breaking into the neuron) and in whole-cell 10 mM EGTA.
Current clamp recordings were performed with the same setting as cell-attached recording, except that pipettes were filled with intracellular solution contains the following (in mM): 120 K-Gluconate, 9 KCl, 10 KOH, 3.48 MgCl2, 4 NaCl, 10 HEPES, 4 Na2ATP, 0.4 NandTP, and 17.5 sucrose, pH 7.25 and Osm 295.
The mean firing frequency of the entire population, measured in each cell immediately after obtaining the cell-attached recording, was 15±15 Hz (n = 76).
Considering that the resting potential of granule cells submitted to cell-attached recording is ∼−65 to −60 mV and the reversal potential for ET-induced current ∼ −25 mV, the ET-induced current steps of 15 pA (see main peak of the distribution, Fig. 7F) may correspond to the opening of single state conductance large pores of over 370 pS, as determined using the Ohm's law.
The decay time constant of the non-negative deconvolution for YC2.60 was ∼1 1.5 s, as estimated from simultaneous cell-attached recording and 2-PI (n = 8 neurons).

At about 10 min after the establishment of the cell-attached configuration, simultaneous loose-seal cell-attached recording and linescan calcium imaging (500 Hz) were performed on the soma of cortical neurons or on the dendrites of Purkinje cells at ˜100 μm from the soma.
There was also considerable similarity between the extracellular potential and the current flowing at the soma, recorded using cell-attached recordings in the voltage-clamp mode (Fig. 2C).
