A vacA s2 allele is more commonly identified with Western strains, and this suggests that European strains were present in northwestern Canada during the ancient individual's time.
Additionally, we investigated whether cytotoxin-associated gene A product (CagA) and vacuolating cytotoxin A (VacA) seropositivity modulated this association.
(The term comes from mad cow disease, though it could also refer to a Brazilian song called "Levada louca" ("crazy rhythm") by Ivete Sangalo, which most of us originally misheard as "A vaca louca" — but I'm digressing here).
The ammonia produced to regulate pH is toxic to epithelial cells, as are biochemicals produced by H. pylori such as proteases, vacuolating cytotoxin A (VacA) [this damages epithelial cells, disrupts tight junctions and causes apoptosis], and certain phospholipases.
Strains of H. pylori that produce high levels of two proteins, vacuolating toxin A (VacA) and the cytotoxin-associated gene A (CagA), appear to cause greater tissue damage than those that produce lower levels or that lack those genes completely. These proteins are directly toxic to cells lining the stomach and signal strongly to the immune system that an invasion is underway.

Main virulence factors of H. pylori for the induction of mucosal inflammation are cytotoxin-associated gene (cag) pathogenicity island- (PAI-) encoded virulence factors, such as cytotoxin-associated antigen (CagA) protein, vacuolating toxin-A (VacA), blood group antigen-binding adhesion (BabA), and outer inflammatory protein (OipA).
In our study, Cag A or VacA mutants can still replicate in BMDC, but were cleared in a fast rate probably due to the interference in the autophagosome formation by Cag A or VacA.
Previously, rhesus macaques were inoculated with the macaque adapted H. pylori strain J166 that contained a s1m1 vacA allele and a functional cag PAI that consisted of two fragments, cag1 through cagB and cagA, separated by 12.5 kb of sequence (Supplementary Fig. 2).
A breakpoint was not determined, but the strain was potentially a true vacA m hybrid as opposed to a dual or mixed infection because of the clean forward and reverse sequencing runs of the two vacA m regions.
The discovery of a similar vacA m region in the Tatshenshini H. pylori strain is significant because a connection has been identified between the ancient strain and local modern Alaskan strains as well as modern Asian strains.
