Binding of STRADα to MO25 involves four interaction sites (sites A D) in STRADα: Glu (site A), Ile (site B), Tyr (site C) and the C-terminal Trp–(Glu)–Phe [W(E F] motif (site D) [ 69, 70].
The first group, from here on named NisA-group, contains a conserved G/A-A/G-x-x-R motif at the C-terminus of the leader peptide and a Ile residue at the first position of the core peptide.

The hydrophobic residue number is in harmony with the number of hydrophobic residues of the protein (chain A: Ile-50, Pro-81, Val-82, Ile-84; chain B: Pro-81, Val-82, Ile-84).
The mutation that we identified at amino acid 145 within motif I changes a Thr, a polar uncharged amino acid, to an Ile, a nonpolar hydrophobic amino acid.
The connection between α-helices V and VI was restored when the conserved Leu195 was replaced by an Ile, a combination that represent the compensated background (Ile195-Ile196, Figure 1D and Figure 1E).
KFC2b could successfully predict only three of them (A-PHE-442, A-ILE-443, and A-LEU-444), whereas PPCheck could predict all the six hotspots (Fig. 5B).
Complex between ATP-dependent HSLU protease ATP-binding subunit HSLU and ATP-dependent protease HSLV (PDB ID – 1G3I; interacting chains – A and G/H): Experimental studies reported six residues, A-ASP-438, A-LEU-439, A-ARG-441, A-PHE-442, A-ILE-443, and A-LEU-444, as hotspots.
(16) In the heme binding NEAT domains of IsdC (IsdC-N1) and IsdH (IsdH-N3), there are an Ile and a Val, respectively, neither of which has the capacity for heme-iron coordination.
The SNP genotyped in our study, rs8192708, is also a missense mutation, causing an Ile to Val change at position 267; however, the functional consequences of this change, if any, are not known.
Thus, M112 was mutated to an Ala, and to an Ile (Fig. 7E) to generate the RNF125 mutant detected in some of the overgrowth syndrome patients31.
