The outcome of A) the 1st round of screening and B) the 2nd round of screening on 152 mutantsnts consisting of 95 tolerant and 57 intolerant M4 lines selected from the first round of mutants.
A total of 496 women (267 BRCandand 229 BRCA2 mutation carriers) completed from 1 to 9 rounds of screening (median 3), for a total of 1847 rounds (Table 1).
Following 12 rounds of screening, we observed a single electrophoretic strip, indicating that the purified ssDNA library with a high affinity for anti-MPT64 antibody was saturated.
Around 7 years later, after approximately 3 rounds of screening in the older group and 4 rounds of screening in the younger, a mortality reduction of 30% was observed and published [ 9], the control group invited to screening and the screening phase of the trial closed.
The results showed the amplified phages from 4 rounds of screening could be well recognized by both Tocilizumab and isotype controls, but no significant difference was observed (Fig. 1B).
We then assumed the introduction of a screening program with 2-yearly screening applied to all women currently 50to6969 years old, at years 1, 3, 5, and so on up to year 19 (that is, 10 rounds of screening in all).
After 5 rounds of such screening, it was found that total phages showed more than 1000-fold selectovitheto tHVGGSSVSSV peptide-coated beads over the control beads (Fig. 1A), indicating the screening successfully enriched some phage clones that selectively bind to the HVGGSSV peptide.
The HPV FOCAL study is well positioned to determine the utility of this proposed approach and to provide evidence for cervical cancer screening guidelines internationally, and further data from completed Round 1 and Round 2 of screening will validate the potential impact of hrhPV on cervical cancer prevention in organised screening programs.
During the initial round of screen, 190 of the 849 RNAi constructs produced anterior touch insensitivity in our hands.
Using the number of positives after five rounds of screens, we estimate that the real false-positive rate of a single round of screen to be [ 190 − (61 − 6 ) ] / 849 = 16 %.
