The mSYCP2 ARLD and mSYCP2 SLD were used as prey.
Similar to control rats (n = 13), SLD (n = 5), PMnR (n = 5), rPCRt (n = 8) and cPCRt (n = 4) lesioned rats exhibited a normal diurnal rhythm of REM sleep, with higher levels during the light period.
We used anti-orexin-B IgG-saporin (OX-SAP) to produce cell-body specific lesions in the rPCRt (rostral to the inferior olive and posterior to the facial nerve) (Fig. 1B), SLD (Fig. 1C), caudal PCRt (cPCRt) (Fig. 1D) and PMnR (Fig. 1E) (see Fig. 2A&D for location of rostral and caudal PCRt).
Additionally, SLD lesions did not affect masseter phasic activity during REM sleep but increased neck muscle phasic twitch frequency (24.16±8.53 twitches/s of total REM sleep in 4 SLD lesioned rats vs 0.98±0.33 twitches/s of total REM sleep in 10 controls, p = 0.0009, Fig. 5).
G1ld, Sld, Slu and G2lu compartments in each DNA histogram of BrdUrd-positive cells were determined manually and numbers of cells in each of these compartments were obtained using statistics option of WinMDI 2.8 software.
For statistical analysis, certain baseline CT parameters (max1D, max2D, max3D, SLD, and volume) were divided into 3 groups: small, medium and large lesions; medium lesions were defined as being within the range of ±20% of the median value.
The proposed method requires that considerable proportions of cells be found in each of the four compartments to be analysed (Slu, G2lu, G1ld, and Sld).

Since the Rad60 SLDs interact with components of the SUMO modification machinery [1], it is perhaps not surprising that ubiquitin cannot substitute for either of the SLDs.
This suggests that sequence similarity may not account for the ability of SUMO to replace the Esc2 SLDs, although it is possible that certain key epitopes in Esc2 SLD2 may be present in S. cerevisiae SUMO, while the same may not be true for Rad60 SLD2 and S. pombe SUMO.
