As discussed in detail (Chih et al., 2001; Chih and Roberts Jr, 2003; Roberts, 2007), utilization of glucose is highly regulated at many steps, whereas lactate utilization is not; lactate use is governed by equilibrative reactions.
These algorithms will alert individuals and counselors to relapse risk based on behavioral decisions that, although seemingly irrelevant at face value, in fact predict a lapse (Bekiroglu et al. 2013; Chih et al. 2014).
It is important to recognize that the low Km of the neuronal transporter, MCT2, not only limits lactate entry, it also restricts lactate efflux from neurons, thereby helping to retain glucose-derived pyruvate for oxidation (Chih et al., 2001; Chih and Roberts Jr, 2003).
Importantly, insertion or skipping of alternative exons at two of the alternatively spliced segments (AS2 and AS4) was reported to regulate binding of neurexins to a number of different interaction partners (Ichtchenko et al., 1995; Sugita et al., 2001; Chih et al., 2006; Koehnke et al., 2010; Siddiqui et al., 2010; Uemura et al., 2010; Reissner et al., 2013).
As a case study we focused on neurexins, a class of synaptic adhesion molecules which are widely expressed in the CNS and play an important role in synapse formation and function (Dean et al., 2003; Missler et al., 2003; Graf et al., 2004; Chih et al., 2006; Taniguchi et al., 2007; Uemura et al., 2010).
Thus, we profiled recognition specificity of neurexin isoforms for three different postsynaptic receptors detected at glutamatergic synapses: neuroligin-1 (NL1B, containing splice insertion B), neuroligin-3 (NL3A2, containing splice insertion A2), and the leucine-rich transmembrane protein 2 (LRRTM2) (Chih et al., 2006; Budreck and Scheiffele, 2007; de Wit et al., 2009; Siddiqui et al., 2010).

The morphological changes in the cells were also examined by staining with Giemsa (Merck, USA) for 10 min and observed under the phase contrast microscope (Nikon H600L microscope), as detailed by Chih et al. [15].
